Cytotoxic Effects of Korean Diospyros kaki (Persimmon) Leaf Extract on HTB-26 and BT-483 Human Breast Cancer Cells
Abstract
Breast cancer poses a major health concern, and recent studies have been directed at examining the potential cytotoxic effect of various natural substances on cancer cells and healthy, noncancerous cells. In the present study, two human breast cancer cell lines, HTB-26 and BT-483, are tested to elucidate the effects of ethanol-extracted Korean Diospyros kaki (persimmon) leaf on human breast cancer cell lines and continue to examine the potential for future clinical use of D. kaki leaf extract as an anticancer agent with fewer adverse effects on normal tissue function. To determine the lowest possible effective dosage, cell lines were exposed to various diluted concentrations of extract and cell's respective media. Following 48 hours of incubation, MTS assays were used to assess cell viability. Results and analysis demonstrate that the extracts displayed cytotoxic effects and statistically significant differences in cell viability in the BT-483 cells between 50 μg/mL and 100 μg/mL and in the HTB-26 cells between 100 μg/mL and 500 μg/mL. Compared to MCF-12A and MCF-12F healthy breast control cell lines, results indicate that persimmon extracts may be effective for naturopathic breast cancer treatment for HR+, HER2+, primary cancers. More research is warranted to explore optimal treatment concentrations and potential for use against HR-, HER2-, metastatic cancers.
